| I. Background | |
| 腫瘤細胞可以借助免疫檢查點(diǎn)受體逃避機體免疫系統的識別和殺傷，因此阻斷免疫檢 查點(diǎn)受體可能是一種廣泛有效的腫瘤免疫治療方法。目前，抗 PD-1/PD-L1 抗體雖然比較 成熟，與抗 CTLA4 抗體類(lèi)似，但由于存在耐藥性，患者的總體有效率較低，因此尋找新 的腫瘤免疫治療靶點(diǎn)迫在眉睫。 Programmed Cell Death Protein 1 (PD-1)，一種在激活的 T 細胞上表達的受體，與其配 體 PD-L1 和 PD-L2 結合，負向調節免疫反應。 PD-1 配體存在于大多數癌癥中，PD-1:PD- L1/2 相互作用會(huì )抑制 T 細胞活性，并使癌細胞逃避免疫監視。 PD1/PDL1 信號轉導通路是 腫瘤免疫抑制的重要組成部分，可以抑制 T 淋巴細胞的興奮，增強腫瘤細胞的免疫耐受， 從而實(shí)現腫瘤免疫逃逸。PD1 與 PDL1 結合可以減弱 T 細胞介導的免疫監視，導致免疫反 應缺失，甚至導致 T 細胞凋亡。PD1/PDL1 抑制劑可解除抗腫瘤 T 細胞的免疫抑制，從而 導致 T 細胞增殖并滲透到腫瘤微環(huán)境中并誘導抗腫瘤反應。PD-1:PD-L1/2 通路還參與調節 自身免疫反應，使這些蛋白質(zhì)成為多種癌癥以及多發(fā)性硬化癥、關(guān)節炎、狼瘡和 I 型糖尿 病的有希望的治療靶點(diǎn)。酪氨酸磷酸酶 SHP2 是 T 細胞功能的關(guān)鍵調節因子，它介導 TCR 下游的激活信號以及 PD-1 下游的抑制信號。 |
